چکیده

Adrenergic receptors are pharmacologically classified into the receptor types 1, 2, 1, 2, and 3. Structural differences and varying affinities in radioligand binding studies lead to a further classification of 1and 2-receptors into subtypes which are termed 1A (formerly 1C), 1B, and 1D (formerly 1AD), and 2AD, 2B, and 2C, respectively. mRNA expression of all but one -adrenergic receptor subtypes and of all -adrenergic receptor types was measured quantitatively in total RNA extracted from mammary tissue of 10 lactating dairy cows by real-time reverse transcription (RT) polymerase chain reaction (PCR). mRNA expression of 1-adrenergic receptors was highest for the 1A-subtype followed by 1B, whereas the 1D-subtype could not be detected. The highest mRNA expression of 2-adrenergic receptors was found for the 2AD-subtype, followed by 2B and 2C. Within the -adrenergic receptors, the 2-receptor type was most highly expressed, followed by 1 and 3. In conclusion, eight of nine adrenergic receptors classified to date were detected and relatively quantified in the mammary gland of dairy cows. © 2002 Elsevier Science Inc. All rights reserved.

Milking characteristics and their relation to adrenergic receptor mRNA expression and ligand binding in the mammary gland of dairy cows.

Stimulation of alpha- and beta-adrenergic receptors in the bovine mammary gland affects milking characteristics such as milk yield and peak flow rate. The aim of this study was to detect possible correlations between milkability, adrenergic receptor binding capacity and receptor expression at the mRNA level. In addition, dose-response relationships of alpha- and beta-adrenergic receptor stimula...

Role of neutrophils in protection of udder from infection in high yielding dairy cows

Protection of the mammary gland against mastitis-causing pathogens is mediated by many factors in thegland and blood circulation. The professional phagocytic cells of bovine udder, polymorphonuclearneutrophils (PMN) and macrophages, comprise the first line of defense against invading mastitis pathogens. Most researchers now accept that the PMN is a key factor in the cows’ defense against intrama...

The bovine mammary gland expresses multiple functional isoforms of serotonin receptors

Recent studies in dairy cows have demonstrated that serotonergic ligands affect milk yield and composition. Correspondingly, serotonin (5-HT) has been demonstrated to be an important local regulator of lactational homeostasis and involution in mouse and human mammary cells. We determined the mRNA expression of bovine 5-HT receptor (HTR) subtypes in bovine mammary tissue (BMT) and used pharmacol...

Short communication: Effect of heat stress during the dry period on gene expression in mammary tissue and peripheral blood mononuclear cells.

Heat stress (HT) during the dry period compromises mammary gland development, decreases future milk production, and impairs the immune status of dairy cows. Our objective was to evaluate the effect of cooling HT cows during the dry period on gene expression of the mammary gland and peripheral blood mononuclear cells (PBMC). Cows were dried off 46 d before their expected calving and assigned to ...
